How avuncular testing helps answer paternity questions

When a paternity test can’t be performed, an avuncular DNA test can provide strong evidence about an individual’s biological relationship to their potential father, by assessing genetic similarities with the potential father’s sibling.

This is possible because:

• Biological siblings (such as the potential father and his full brother or sister) share around 50% of their DNA, inherited from their parents.
• If the potential father is biologically related to the tested individual, the tested individual will also share a portion of DNA with the father’s biological sibling(s) – typically around 25%.
• By analyzing these shared genetic markers, scientists can calculate the probability that the potential aunt or uncle is biologically related to the tested individual

This makes avuncular testing particularly useful in circumstances where the potential father is:

• Unreachable or unwilling to take a DNA test
• Deceased
• Not named on the birth certificate, and paternity needs to be determined through other paternal relatives

The result of the test is not a simple “yes” or “no” answer, but a likelihood ratio – a statistical estimate showing how probable it is that the tested person is related to the tested individual in the stated way.

Due to the nature of avuncular DNA testing, we recommend that the potential aunt/uncle who has a claimed biological link to the child/adult should be tested along with the biological mother, if this is an option. However, the test can still be performed without a biological parent’s DNA.

When is an avuncular test recommended?

Avuncular DNA testing is most commonly recommended when a paternity test is not possible, and there is a need to establish whether an individual is biologically related to the potential father’s side of the family.

• The potential father is unavailable or unwilling to participate – in cases where he cannot or will not provide a sample.

• Under 10% = unlikely to be related
• Between 10% and 90% = inconclusive, meaning a relationship cannot be statistically supported by the analysis

Avuncular testing for maternity

While avuncular testing is most often used to help determine paternity, it can also be used to confirm maternity where a standard maternity test is not possible (e.g. the biological mother is deceased or unavailable).

For an avuncular test for maternity, a full biological sibling of the potential mother would need to provide a DNA sample for testing, along with the potential niece/nephew.

Although the need to confirm maternity is less common, there are several circumstances where avuncular testing for maternity might be needed, including:

• Adoption or long-term separation – where a person wants to confirm whether a woman is/was their biological mother, but direct testing is not possible.
• Missing persons or historical investigations – in situations involving unknown parentage or unregistered births, particularly where traditional records are lacking.
